Nanopore 16S Amplicon Sequencing Enhances the Understanding of Pathogens in Medically Intractable Diabetic Foot Infections

Diabetic foot infections (DFIs) cause substantial morbidity and mortality. The mainstay of the treatment is empiric antibiotics surgical debridement in severe cases. In this study, we performed nanopore 16S rDNA sequencing from specimens DFIs. Fifty-four obtained 45 patients with medically intractable DFI were included. PCR was on each specimen, Nanopore for up to 3 h. reads aligned BLAST database, results compared conventional culture studies. revealed that majority DFIs (44 54, 81.5%) polymicrobial infections. All bacteria isolated by studies detected sequencing. Several anaerobes (Prevotella, Finegoldia, Anaerococcus, Bacteroides) commonly identified but frequently missed many cases, certain only more abundant than conclusion, capable pathogen identification has advantages over enables a comprehensive understanding involved
